Once your protein has been purified, we can determine the size, purity, concentration of your protein using, SDS-PAGE, western blot, as well as other protein quantitation techniques (spectrophotometer for UV-Vis, BCA and Bradford, nanodrop, Direct Detect).
For more specific characterisation tests, the RPF has:
- An Agilent Bioinert HPLC
At the Recombinant Products Facility, we come across very different projects requiring the use of a large variety of HPLC columns and methods.
Our Bioinert HPLC with its temperature regulated autosampler (from 4 to 40C), DAD fluorescence detector and fraction collector in combination with our wide range of HPLC columns available is a great asset to analyse your protein; for example to determine protein homogenity (charged variants, glycosylation) or aggregation.
We have a selection of Size exclusion, anion, cation exchange as well as Reverse Phase columns (list available on demand). Some clients prefer to bring their own specific columns, and we develop for them a method for their own characterisation.
- Prometheus (nanotemper)
The Prometheus NT48 allows the measurement of protein unfolding and aggregation.
Buffer formulations can be screened to find the optimal buffer of your final product.
This instrument is as well used as quality control for:
-Long-term stability of biologics
-Forced degradation
-Thermal stability of antibody/Drug conjugates
and has a specific software developed for stability studies.
UNSW hosts other accessible facilities (see UNSW Research Infrastructure ) Listed below are those specifically related to proteins.
